H3G34-mutant hemispheric gliomas exhibit constitutively elevated H3K36me3 (trimethylation at H3 lysine 36), which disrupts normal chromatin architecture and represses genes normally silenced by H3K27me3, creating an epigenetic state intermediate between wildtype and H3K27M-mutant tumors.
